The subject here is rather large and without good lighting. You need a bigger depth of field- you are at f/4.0. I would have increased the f as much as possible and allowed the iso to go as high as it works well without a lot of noise on your camera. (your camera goes to 12800, so you could probably go to 3200 without a lot of noise). But obviously, sometimes the shutter speed can be slower than you like for a poorly lit subject. Thus, I feel that really good IS (image stabilization) is crucial in a macro lens.
