Theres a building you can go up on the eastern side of the Charles Bridge looking towards the castle, can't remember what its called but great photo spot.
agree about charles bridge and the castle. Also :
- there may be something on the bus / train ride up to the castle
- Old Town Hall / astronomical clock and meandering around Old square
- Letna Park, bring your wide angle lens, a tripod and shoot the view of the city. Metronome may be interesting too. I got engaged here :)
- Views down Wenceslas square
- Slovansky / Detsky etc islands just in the river. Nighttime shots here also
- Synagogues
- Jewish Cemetary
- If you want something really amazing, book dinner at a restaurant called Svata Klara. Its just out of the city (get a cab and get your hotel to send a cab to pick you up). Its a restaurant in the underground cellar of an old winery, absoltuely amazing. Bring your tripod or a flash lol
@limpet365 : The whole city is a paradise for photographers! A view from the Charles Bridge is standard of course but I especially loved the museum of modern art and the old train station.

I agree on Charles Bridge and castle and add the stained glass windows in the church at the castle, and awesome art nouveau architecture - from memory some great examples near Wenceslas square, gorgeous lane ways in Stare Mesto (old town) @jannaellen is spot on photographers paradise
My favorites were the Old Town Square and Prague Castle. The Charles Bridge is ok, but it's just nearly impossible to photograph due to the quantities of people massing on it at all times. Your best bet is to head south a bridge or two, then you can actually get photographs with the bridge itself in it.
If you can swing it, climb the Astronomical Tower right before it closes. They let you stay up as long as you like, and we got some simply incredible sunset pictures. This was my 365 pic for that day ...
If you click around a few days before and after that shot, you can see some other views. Also, if you like lines and patterns, be sure to go down and check out the Soviet-style subway stations!
Don't forget the wonderfully ornate and decorative architecture! Everywhere you look there are frescoes, carvings, sculptures, paintings, verdigris roofs and domes. But make sure you look UP as well as around because so much of it is several floors up and easy to miss at street level.
